The most famous for his role as a stoner would definately be Sean Penn. His role as Spikoli in Fast Times at Ridgemont High sparked his career, and made him one of the best actors of our times. After this break he went on to play multiple different roles. He was a cop in Colors, a mentally challenged father in I Am Sam, and his award winning role of an ex-thug father in Mystic River.
The next actor is a really famous actor now, that got big by playing a stoner in the classic movie Easy Rider. His name is Jack Nicholson and he has been nominated for many oscars. The roles I best know this man from are the father in The Shining, a werewolf layer in Wolf, and even as the funny and strange therepist in Anger Management. He has played many other roles, but the most important thing is that he got famous by playing a stoner.
The next actor got known through Thelma and Louise, but didn't become famous till after playing a stoner in True Romance. This actor is Brad Pitt and he has had two really big hits as a cop in Seven and a badass soap salesman in Fight Club. He has been in multiple other films and even hooked up with Jennifer Aniston and Angelina Jolie.
This actor isn't really well known anymore, but for a while in the nineties he was a pretty big comedy star after playing a stoner named Smokey in Friday. His name is Chris Tucker and after this movie it seemed like he was going to be a big star in comedy for years to come. After his role as Smokey he played a hustler in Money Talks and a smart mouth cop in the Rush Hour series. After Rush Hour 3 I haven't heard much about him since.
The final actor is one that just got his break playing a drug dealer named Saul in Pineapple Express. I believe that he stole the show with this portrayal. The actor I'm speaking of is James Franco. He has played roles as a rich best friend in Spiderman, a criminal son in City by the Sea, and Sean Penn's lover in Milk. The movie Milk has not only one but two actors made famous by their portrayals of a stoner and was up for many awards.

It's cool that you are bringing some focus to the public's fascination with people playing stoners. It's nice to see another side to your quest, that some stereotypes arn't always seen as bad in certain perspectives.
